Restriction digests of the clone give the following sizes (kb): BamHI--4.4, 1.0; HindIII--4.6, 0.8; PstI--4.2, 1.2; PvuII--3.2, 2.4. The 1.0 kb (approx) BamHI fragment includes the cDNA of 0.93 kb which contains the complete coding region plus 5' and 3' untranslated sequences. The BamHI fragment also includes, at the 5' end, at least 102 bp of the original pcD vector. The BamHI fragment of pcD-mut-1 was cloned into the BamHI site of pBR322. There is also pcD vector sequence at the 3' end. |
