SphI, KpnI, XbaI, XhoI, SalI and BamHI sites are probably unique within the insert. Insert contains the following sites separated by (kb) (approximate): EcoRI- 0.58- BglII- 0.2- BglII- 0.2- SphI- 1.1- XhoI- 0.43- KpnI- 1.5- PstI- 0.22- SalI- 1.26- BamHI- 1.53- EcoRI. Approximate sizes of fragments (kb)(from depositor): EcoRI--7.0, 3.2; EcoRI+KpnI--4.4, 3.2, 2.6; EcoRI+BamHI--5.5, 3.2, 1.5; EcoRI+SalI--4.2, 3.2, 2.8; EcoRI+XhoI--4.9, 3.2, 2.1; EcoRI+XbaI--4.5, 3.2, 2.5. Restriction digests of the clone give the following sizes (kb): EcoRI--8.4, 3.6; BamHI--12.2; SalI--12.2; EcoRI/BamHI--6.4, 3.6, 1.8; EcoRI/SalI--5.0, 3.6, 3.2. The KpnI site is located in the first exon just 5' of the first AUG (position +24). More homology between mouse and rat WAP genes in 3' noncoding (91%) than coding (82%) regions. For 5' region, homology between mouse and rat genes is 83% and extends at least to position -325. |
