Animals were immunized with a mixture of six prostate tumor cell lines: primary cell lines, ALVA31, JCA1 and ND1; metastatic cell lines, DU145, LNCaP and PC3. 5E10 recognizes an antigenic determinant on the surface of normal and cancerous prostate cells but not on the surface of non-prostatic tumor cells, peripheral blood cells, bone marrow hematopoietic and stromal fibroblast cells, or endothelial cells. The epitope recognized by 5E10 is extracellular. Western blot analysis demonstrates the 5E10 antigen is a transmembrane glycoprotein that has a molecular weight of about 115 Kd. A culture deposited at the ATCC as PTA-865 in October of 1999 was found to be contaminated with mycoplasma. Progeny were cured by a 21-day treatment with BM Cycline.
