Recommended for use in the tests described in Fed. Spec. A-A-1438 where only the taxon is specified.
This item was originially adapted for growth in CE tissue. Pocks on CAM tend to be larger than those produced by other human herpesvirus 1 strains. Large inocula are required to infect animals.
Vials contain total fluid and material from cell culture infected by virus. The vials will contain cellular debris and possibly cells containing cell-associated virus. |
| Effect on Host |
CPE, syncytia, rounding, detachment |
| Recommended Host |
Vero (ATCC CCL-81) |
| Growth Conditions |
Temperature: 37°C
Recommendations for Infection: Plate cells 24 to 48 hours prior to infection and infect when cultures are 80-90% confluent. Remove medium and inoculate with a small volume of virus (e.g. 1 mL per 25 cm2) diluted to provide an optimal MOI (e.g. 0.1). Adsorb 1-2 hours at 37°C in a humidified 5% CO2 atmosphere, rocking every 20 to 30 minutes to redistribute inoculum. End adsorption by adding virus growth medium.
Incubation: 2-3 days |
