The insert contains internal EcoRV, BamHI, AccI and HincII sites. Restriction digests of the clone give the following sizes (kb): SmaI--uncut; BamHI--4.0, 1.8; EcoRI--5.6; HindIII--5.6; PstI--5.6; EcoRI/HindIII--3.0, 2.7. Linearize the plasmid with HindIII for in vitro transcription. This is a full-length clone of virus segment S5. The cDNA was tailored to have a 3'-terminal EcoRV site before cloning into the SmaI site of pGEM-1. |
