The insert includes the following restriction sites (in nucleotides from the 5' end of the published sequence): SmaI--319, 580; SphI--440, 1649; PstI--638, 2780, 3256; SacI--1201; XhoI--1763. Restriction digests of the clone give the following sizes (kb): EcoRI--3.3, 3.0; HindIII--5.1, 1.35; BamHI--6.4; PstI--3.6, 2.2, 0.59; SalI--4.1, 2.2. The sequence was confirmed by partial sequence analysis and it includes and matches nucleotides 250 to 3347 of the published sequence. It encodes the complete coding region (437 - 1841) plus some 5' and 3' untranslated regions. A 1.209 kb SphI fragment of the probe was used to detect a genomic PvuII fragment of 7.1 kb in the rat and a genomic PvuII fragment of 3.7 kb in the mouse. The insert is oriented such that linearizing with BamHI and using the T3 promoter gives the sense strand, and linearizing with EcoRI and using the T7 promoter gives the antisense strand. |
