Restriction digests of the clone give the following sizes (kb): PstI--2.8, 0.5; BamHI--3.4; EcoRI--3.4. This clone is useful in hybridization tests for the detection of tobacco streak virus (necrotic shock) infection in Fragaria spp. Insert hybridizes with NS (necrotic shock) RNA 3 from strawberry and R RNA 3 from blackberry but not to R RNA 4 in northern blots of genomic RNA. Isolated from Fragaria vesca with strawberry necrotic shock. cDNA was synthesized from virion genomic RNA, dC-tailed and cloned into the dG-tailed PstI site of pUC8. |
