Restriction digests of the clone give the following sizes (kb): EcoRI--3.0, 2.2; HindIII--5.2; PstI--4.3, 0.9; BglI--2.0 (doublet), 1.2; SalI/XbaI--3.1, 1.15, 0.5 (doublet). The 5' clone phgJE3-3 (ATCC 61366/61367) and the 3' clone phgJE2-1 (ATCC 61368/61369) together generate a complete gene. The insert is oriented such that the EcoRI site in intron 1 is closer to the T7 promoter. |
