Reported to contain EcoRI/HindIII fragments of the following sizes (kb), ordered as in the genome: 4.762, 0.408, 0.702, 0.902. Restriction digests of the clone give the following sizes (kb): EcoRI--9.0, 6.8; HindIII--9.8, 4.8, 0.7, 0.4; EcoRI/HindIII--9.0, 4.8, 0.9, 0.7, 0.4; PvuI--12.0, 3.2. The first 6.6 kb of the construct measured from the YIp5/yeast DNA junction contains the following restriction sites (kb from the HindIII site): PvuI--2.3; BamHI--4.0; HindIII--4.7, 5.1, 5.8; EcoRI--6.6. Overlaps clones with ATCC number(s): 70867. The RAD4 gene is contained within the 4.8 kb EcoRI/HindIII fragment. Isolated after plasmid rescue as an intramolecular ligation of a SalI fragment. Consists of YIp5 from SalI to the HindIII cloning site of the original integrant pR169, spt2 starting at the HindIII site, and the complete RAD4 gene. |
