Restriction digests of the clone give the following sizes (kb): HindIII---4.0, 2.3; EcoRI--6.3; PstI--4.5, 1.8; BamHI--4.7, 1.8; SalI--6.3. This probe codes for 182 Gly-Xaa-Yaa residues, the 229 amino acid COOH-terminal noncollagenous peptide and 435 nucleotides of the 3' untranslated region. This insert includes the 5' portion of the 3' untranslated region and contains only one of the many AATAAA polyadenylation signals of the mRNA. The collagenous region is encoded primarily in the 5' PstI-BamHI (0.52 kb) fragment of the insert. The non-collagenous carboxyl terminus (NC1) is encoded in the adjacent BamHI-HincII fragment (about 0.72 kb). The HincII-PstI fragment contains the 3' untranslated sequences. This probe detects mRNA of 6.5 kb (major) and 5.9 and 5.3 kb (minor) in human skin fibroblasts. |
