The following enzymes cut the insert once (in nucleotides from the 5' end of the insert): XhoI--209; PvuII--372; SalI--481; HindIII--2410; PstI--2493; BamHI--2774. Restriction digests of the clone give the following sizes (kb): EcoRI--3.2, 2.9; HindIII--5.3, 0.78; PstI--3.9, 2.1; XbaI--6.1; HincII--3.4, 1.4, 1.3. This clone encodes a portion of the extracellular region and the complete transmembrane and tyrosine kinase catalytic domains. It includes a 1376 bp 3' non-coding region. There are no consensus sequences for polyadenylation near the 3' terminus, but there are 4 ATTT(A) motifs which have previously been identified in lymphokine and oncogene mRNA and which have been associated with message instability. This probe detects genomic BamHI fragments of 4.6, 3.3, and 1.5 kb in humans. The probe detects diverse mRNA species which are tissue-specific and developmentally regulated. Sizes are given in kb. Brain: 9.0, 8.2, 8.0, 5.5, 3.5, 2.5, 2.0; lung and muscle: 8.2, 8.0, 3.5, 2.5, 2.0; ovary: 8.2, 2.5; testis: 2.8, 2.5. |
