Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--2.7, 1.2, 0.9; BamHI--4.8; PstI--2.9, 1.9. The plasminogen portion corresponds to nt 196-697 of the reported sequence and encodes the K1 kringle domain and part of the K2 kringle domain. Downstream of the plasminogen sequence is a vector sequence encoding the peptide GGSSR and a truncated streptokinase gene lacking regulatory sequences and codons 1-39 but retaining the remainder of the coding sequence and the transcription terminator. Contains 2 internal PstI sites. Encodes a plasminogen-streptokinase fusion protein expressed under control of lacPO. The fusion protein is extended at the N-terminus by the N-terminal hexapeptide of LacZ' followed by four polylinker-encoded amino acids. The plasminogen sequence was cloned into the SmaI site and the streptokinase sequence was cloned into the HincII site. |
