Restriction digests of the clone give the following sizes (kb): SalI--5.72; PstI--5.72; HindIII--5.68, 0.4; EcoRI--5.6, 0.65; AvaI--5.72. IMPORTANT: To prevent amplification of a rearranged and/or deleted cosmid, we recommend streaking on LB + amp plates at 30C and picking small colonies for liquid culture. The BamHI RFLP is also detected by p13-1-82 (ATCC 59004, 59005). The combined heterozygosity of the polymorphisms detected by p13-1-82 (ATCC 59004, 59005) and p13-2-21 (ATCC 59014, 59015) is 72%. Seven of 24 possible haplotypes occur with frequencies of 3-38%. p13-1-82 (ATCC 59004, 59005), p13-2-31 (ATCC 59014, 59015), and p13-1-25 (ATCC 59870, 59871) were subcloned from cosmid 1-13 (C1-13). |
