Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--2.7, 2.3. An EcoRI/HindIII double digest shows an insert fragment size of 2.3 kb. This plasmid shows some instability. We recommend growing on LB agar with ampicillin, and picking colonies to select the correct clone. Codominant segregation of the MspI polymorphism has been demonstrated in 45 3-generation families.
