Restriction digests of the clone give the following sizes (kb): BamHI/SalI--6.0(doublet); EcoRI--12.0; SmaI--12.0; Pvu/BamHI--6.5, 2.5, 2.1, 0.54, 0.49. Expresses beta-galactosidase activity in E. coli, S. cerevisiae, and mouse cells, and thus can be used as a control to calibrate transformation efficiency. Shuttle reporter plasmid. Constructed by inserting a BamHI/SalI fragment from pMC1403 (lac operon beginning at codon 9 of lacZ) between the BamHI/BalI (SalI-linkered) sites of pCYM0. Our plasmid is 12 kb instead of 9.4, which matches the original depositor's DNA. |
