Restriction digests of the clone give the following sizes (kb): XbaI--4.5, 1.4; HindIII--3.8, 1.3, 0.80; PstI--5.9. pEA2 carries his7+ cloned into the pUC18 polylinker. To construct gene disruptions from this vector, fragments carrying his7+ can be isolated using a variety of restriction enzymes. Constructed by inserting the 3.15 kb BglII fragment from plasmid pMN1 into the BamHI site of pUC18. |
