Restriction digests of the clone give the following sizes (kb): BglII--5.0, 2.7, 0.62; PstI--3.3, 2.7, 2.4; PvuII--6.5, 1.7. The insert contains the following restriction sites (approximate kb from the 5' end): BglII--0.0, 0.6; EcoRV--1.2, 2.3; PstI--0.1, 2.4; PvuII--1.5; SacI--0.7. A partial cDNA clone corresponding to nt 1-3138 of the reported sequence (complete coding region 179-3472). The truncated product still confers LIF binding activity. Detects transcripts of approximately 6 kb, 4.5 kb and 5 kb (minor) in human placenta. pDC302 is a mammalian expression vector containing promoter and enhancer sequences from SV40 and CMV, leader sequences from adenovirus 2 followed by a multiple cloning site, SV40 polyadenylation and termination signals and adenovirus genes VAI and VAII. Digestion with BglII yields a 5 kb vector fragment and fragments of 2.6 kb and 0.60 kb from the insert. |
