Restriction digests of the clone give the following sizes (kb): XbaI/SacI--2.9, 1.1; EcoRI--4.0; HindIII--4.0; XbaI--4.0; SacI--4.0. pGEM-3.110B-1 has been deposited in the NIH repository (ATCC 61328) and to support a patent application (ATCC 67566). This clone encodes a complete alpha chain and includes 5' and 3' flanking sequences with a poly(A) tail. The XbaI site is at the 5' end. The probe used to detect the RFLP was a 0.9 kb XbaI/SfaN1 fragment which includes the entire reading frame but excludes some of the 3' sequences. The entire cDNA was also used. The probe detects human EcoRI fragments of 4.4, 3.0, and 1.5 kb and mouse EcoRI fragments of 4.9, 3.9, and 1.7 kb. Enzyme(s) not detecting polymorphism: AccI, AvaII, BamHI, BanI, BglII, DdeI, EcoRI, EcoRV, HincII, HindIII, HinfI, MspI, PstI, PvuII, SspI, TaqI, XbaI. |
