Insertion at the HindIII site makes the plasmid unstable. Insertion at the BglII (1.5 kb) or BamHI sites destroys plasmid self-transmissibility.
Insertion at the BclI (0.3 kb), BclI (4.5 kb), BglII (6.1 kb), or BglII (7.2 kb) does not affect plasmid self-transmissibility.
Has demonstrated ability to be maintained in S. Venezuelae, S. Lividans, and S. Avermitilis.
