Restriction digests of the clone give the following sizes (kb): HindIII--4.4, 2.5; PstI--4.4, 2.5; EcoRI--5.6, 1.35. Encodes a catalytically functional enzyme (aa 1-337). Deletion of the C-terminal 49 aa desensitizes the enzyme to phenylalanine feedback inhibition. Excision of the insert with HindIII yields a lac promoter - pheA' - aroF cassette. Production of phenylalanine is enhanced when the pheA structural gene is introduced as part of a synthetic operon, rather than as a single deregulated structural gene. |
