Restriction digests of pNEB671 give the following sizes (kb): BamHI--10.5; BamHI/NdeI--6.2, 2.1 doublet; EcoRI--9.0, 1.9; HindIII--9.0, 0.7, 0.7. pLysS is also detected at 4.9 kb (or 4.2 and 0.6 kb with BamHI).
Constructed by modifying the sequence TTTATG to CATATG to create a NdeI site at the 5' end (nt 288-293), and by removing the intervening sequence spanning nucleotides 1776 and 3398 of the sequence record.
