The GAP/xylA/xylB operon may be recovered as a 4.1 kb NotI fragment.
This strain containing pZB4 is useful for fermenting glucose and pentoses (xylose), produced by hydrolysis of cellulose and hemicellulose, to produce ethanol.
The xylA/xylB operon is controlled by the Z. mobilis glyceraldehyde-3-phosphate dehydrogenase (GAP) promoter.
The talB/tktA operon is controlled by the Z. mobilis enolase (ENO) promoter. The GAP and ENO promoters are in a head-to-head orientation.
Plasmid contains the GAP-xylA/xylB operon from pGapXKH in a clockwise orientation and ENO-tal/tkt operon from EnoTAL/TKT in a counterclockwise orientation.
This plasmid is shipped in Zymomonas mobilis CP4. It should be grown in Zymomonas media (20 g glucose, 10 g Yeast extract, 2 g KH2PO4 in one liter of distilled water) with 10 ug/ml tetracycline at 30C in an anaerobic gas mixture of 80%N2-10%CO2 - 10% H2.
