Comments
The construct contains a full-length Human Rhinovirus type 14 (HRV14) genome cloned into the StuI site of the pMJ3 vector.
HRV14 cDNA was transplanted into the vector at the StuI site.
The 5' end of the cDNA is two bases (GG) from the transcription initiation site of the T7 promoter. A unique MluI site (ACGCGT) was added after the polyA tail. Full length transcripts can be produced using T7 RNA polymerase. T7 transcripts are half as infective as genomic RNA from virons.
