Restriction digests of the clone give the following sizes (kb): EcoRI--7.3; BamHI--7.3; HindIII--7.3; PstI--4.4, 2.7; EcoRI/HindIII--4.4, 2.7. This clone is difficult to maintain, with deletions and/or viability loss with prolonged storage on LB agar. The depositor recommends storing as DNA. The fragment begins at nt 1285 of the HXP sequence (EMBL X13993). Downstream from the HXP sequence there is about a 3.0 kb long unsequenced gap with no known restriction sites, then the fragment continues and ends with the XXP sequence (EMBL X13991). This PstI fragment includes a 3.1 kb PstI/XbaI fragment with no known restriction sites. The 5' portion of the 3.1 sequence is A-rich; followed by a GC-rich region, and a 3' pyrimidine-rich sequence composed mainly of CT dinucleotides. The 4.3 kb XbaI repeat occurs in 2 copies in a 10.3 kb HindIII fragment (HeLa cells) of the non-transcribed spacer of ribosomal RNA. The insert contains Alu type repeats. |
