Restriction digests of the clone give the following sizes (kb): BamHI--17.0, 7.4, 3.5, 3.2, 2.9; BglII--13.0, 7.8, 6.6 (doublet), 2.4, 2.1, 1.7, 1.55, 0.85, 0.7; HindIII--9.4, 6.6 (doublet), 5.6, 5.0, 4.0, 2.6, 2.05, 1.5, 0.9, 0.65; PvuII--11.0, 5.4, 4.2, 3.0, 2.3, 2.15, 2.05, 1.55, 1.1, 0.9, 0.7; SacI--10.0, 9.4, 5.8, 3.8, 3.1, 2.2, 1.8, 1.65, 1.45, 1.3, 1.0, 0.75; KpnI--20.0, 8.4, 7.8, 3.1. IMPORTANT: To prevent amplification of a rearranged and/or deleted cosmid, we recommend streaking on LB + amp plates at 30C and picking small colonies for liquid culture. Contains the complete gene which spans approximately 18 kb and includes 10 exons. GenBank M59856 records 5' flanking sequence, exon 1, and part of intron 1. GenBank M59783 begins about 2.3 kb downstream and includes the rest of the gene. |
