Restriction digests of the clone give the following sizes (kb): 18.0 + arms. This probe has been assigned to a linkage group on 16q (Lathrop et al., HGM9). The order of marker loci is cen-HP-CTRB-D16S20-pLC9-(APRT,D16S7)-qter. Highly polymorphic locus with RFLPs observed with BclII, BglI, BglII, BstNI, MspI, PstI, PvuII, SacI and TaqI. Most seem to be site mutations. Sites A, B, and D show a heterozygosity of 86% (sample of 36 unrelated individuals). The whole phage works well as a probe after prehybridization with total human DNA to block annealing of repetitive sequences. The final stringency wash is 0.1X SSC/ 0.1% SDS at 65C. |
