Restriction digests of the clone give the following sizes (kb): EcoRI/KpnI--2.4, 1.0; BglI--3.4; PstI--3.4. Encodes a catalytically functional enzyme (aa 1-337). Deletion of the C-terminal 49 aa desensitizes the enzyme to phenylalanine feedback inhibition. Constructed from pKB750 (ATCC 39857) by subcloning the pheA gene fragment into a pKB430 derivative and then deleting the NcoI/EcoRI fragment of the insert. |
