Restriction digests of the clone give the following sizes (kb): EcoRI--9.0; BamHI--9.0; EcoRI/BamHI--4.75, 4.1; HindIII--uncut; PstI--3.3, 1.4, 1.3, 1.05 + others. This insert contains the complete zeta globin pseudogene 1 with approximately 1.5 kb flanking sequence upstream and 1.1 kb flanking sequence downstream. A 1.16 kb BglII/PvuII fragment which includes parts of exon 1 and intron 1 (IVS-1 probe) has been used to detect the SacI and PvuII polymorphisms. A 1.8 kb SacI fragment has been used to detect XbaI and PvuII polymorphisms. The 5' EcoRI end of the insert was engineered during construction of the probe. |
