Animals were immunized with TC-31 (ATCC CRL-2494), a monoclonal antibody to exotoxin A of Pseudomonas aeruginosa. Spleen cells were fused with Sp2/0-Ag14 myeloma cells. The antibody binds specifically to an idiotypic determinant on TC-31 anti-exotoxin A monoclonal antibody. Exotoxin A inhibits the binding of the 9-2-4 antibody to TC-31. This antibody conjugated to keyhole limpet hemocyanin (KLH) can induce antibodies in mice that bind to exotoxin A and protect 80% of the mice from a lethal injection of exotoxin A. |
