Restriction digests of the clone give the following sizes (kb): SacI--3.9; SalI--2.8, 1.1; XbaI--2.3, 1.4.
The insert contains the following restriction sites (approximate kb from the 5' end): BglI--0.18, 0.37; SalI--0.34; SacI--1.11.
Digestion with XbaI yields an intact insert with a portion of the T7 phi 10 promoter included at the 5' end and a portion of the MCS at the 3' end. Extraneous 3' material can be removed with an EcoRI digest.
The pH 2.5 acid phosphatase is more active as a phytase.
