Colonies of different sizes may be observed. Region specific library constructed by microdissection of metaphase chromosomes, complete digestion with MboI, followed by PCR amplification and subcloning of amplification products. Approximately 80% of unique sequences were derived from chromosome 2, as determined by hybridization. Half of all the clones have repetitive sequences and all repetitive sequences should be human and chromosome 2 specific. The library is estimated to be 90% pure. Approximately half of the clones in the library contain unique or low-copy number inserts. The mean insert size is 190 bp. |
