Restriction digests of the clone give the following sizes (kb): BamHI/HindIII--5.8, 0.75, 0.6; HindIII--6.6, 0.6. A 0.6 kb HindIII fragment contains the coding sequence plus some 3' untranslated sequence. This should not be grown at temperatures exceeding 30C. This was constructed from pAW711 (ATCC 39918) by deleting nucleotides encoding 6 amino terminal amino acids starting with Val of the processed TNF. This product shows maximal biological activity. |
