Restriction digests of the clone give the following sizes (kb): BamHI/EcoRI - 6.2, 2.8; BglI--2.4, 2.0, 1.4, 1.15, 0.95, 0.5, 0.3, 0.27; HindIII--3.8, 3.4, 1.65, 0.25. Expression of both enzyme activities is nearly optimal for both anaerobic and aerobic growth. The GenBank accession number for the adh insert is M15394. Constructed by inserting a promoter-less BamHI/EcoRI fragment from pLOI293 into the vector, and then inserting a promoter from Zymomonas mobilis into the BamHI site which is immediately proximal to the pdc gene. |
