Restriction digests of the clone give the following sizes (kb): BamHI--3.0; EcoRI--3.0; HindIII--3.0. High-copy-number plasmid (in bacteria) with increased efficiency of liposome-mediated introduction into cultured cells. Can express foreign sequences in cells producing T7 RNA polymerase, such as that encoded by recombinant vaccinia virus vTF7-3 ATCC VR-2153. A correction to the published sequence has been submitted. Sequence 2173-2252 should read: GCGACCGAGT TGCTCTTGCC CGGCGTCAAT ACGGGATAAT ACCGCGCCAC ATAGCAGAAC TTTAAAAGTG CTCATCATTG. Constructed by inserting the 152 bp BamHI-BglII fragment from pAR2529 into the BamHI site of pTZ18R. Polylinker sites were removed and were replaced with the 48 bp EcoRI-KpnI fragment of pBluescript KS+. The order of the major features of the plasmid is: universal primer sequence - XbaI - T7 phi 10 terminator < - XbaI - BamHI/MCS/EcoRI - < T7 phi 10 promoter - reverse primer sequence - pMB1 ori - < ampR - f1 ori. |
