CpEgt1、MsEgtA和MsEgtE對釀酒酵母進行密碼子優化，而其他基因使用GeneArt提供的密碼子優化工具對釀酒酵母進行密碼子優化。所有酵母菌菌株的構建均采用CRISPR/Cas9和易克隆-無標記方法，將一個額外的NcEgt1和/或CpEgt2表達式拷貝整合到ST8461中。
