Applications

The SWLA1 (ATCC HB-12559), SWLA2 (ATCC HB-12580), and SWLA3 (ATCC HB-12558) antibodies appear to recognize a species-specific lipooligosaccharide or lipopolysaccharide on the cell surface of S. mutans.

These antibodies can be used in the clinical diagnosis and treatment of dental caries in humans.

Animals were immunized with Streptococcus mutans Clarke (ATCC 25175).

Spleen cells were fused with NS-1-Ag4-1 myeloma cells.

These antibodies have substantially no reactivity with any of the following bacterial strains: Streptococcus rattus (ATCC 19645, Streptococcus gordonii (ATCC 10558), Streptococcus gordonii (ATCC 13396), Streptococcus mitis (ATCC 49456).

Complete Growth Medium

HL-1 medium supplemented with 4 mM L-glutamine, 1 mM sodium pyruvate, 10 mM HEPES and 10% fetal bovine serum. HL-1 medium can be obtained from Lonza (catalog number 77201).

Subculturing

Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 1 to 2 X 10 exp5 viable cells/ml.

Maintain cell density between 1 X 10 exp5 and 1 X 10 exp6 viable cells/ml.

Note: About 50% of the cells may attach. Cells in suspension grow singly and in small clusters.
