The oriT-containing fragment can be excised more precisely with an EcoRI/BamHI double digest. The NotI digest excises a fragment of approximately 1.9 kb that includes short segments of the pHSS21 vector.

The NotI cassette includes these features: pHSS21 sequence - EcoRI - oriT - PstI - BamHI - XbaI - SalI - PstI - SphI - HindIII - pHSS21 sequence.
