Restriction digests of the clone give the following sizes (kb): XbaI--7.2, 0.3; PstI--3.4, 3.25, 0.95; HindIII--7.5. pVE328 is available as ATCC 67698 and ATCC 68408. IMPORTANT: To prevent amplification of a rearranged and/or deleted cosmid, we recommend streaking on LB + amp plates at 30C and picking small colonies for liquid culture. Cosmid shuttle vector permitting packaging both as bacteriophage lambda and TG1 virions. The order of the major features in this plasmid is: pVE1 ori - BalI - tsrR - SstI - lambda cos - lamda cos - HindIII - TG1 cos - XbaI/MCS/PstI. |
