Restriction digests of the clone give the following sizes (kb): BamHI--3.25, 0.7; PstI--2.55, 1.2; XhoI--3.15, 0.76; SalI--4.0; HindIII--4.0. Codominant segregation demonstrated in 2 informative families with 10 individuals. Digest with HindIII gives the following constant bands: 15, 9.4, 7.2, 4.6, 2.45, 1.35, 1.25, 1.05. EcoRI detects 10 invariant bands and an additional polymorphic band at 1.8 kb (in 10/53 persons). Only insert DNA should be used for nick translation. Enzyme(s) not detecting polymorphism: BamHI, BglII, MspI, XbaI using a panel of 9 unrelated Caucasians. |
