Restriction digests of the clone give the following sizes (kb): NcoI/XbaI--4.0, 0.5; HindIII/NcoI--4.0, 0.5; EcoRI--4.6. Contains part of the hinge region of immunoglobulin gamma 1. The heavy chain and light chain variable regions may be cloned into the PstI-EcoRI-BstEII and SstI-PmlI-XhoI multiple cloning sites respectively. The other unique restriction sites (XbaI NcoI HindIII) may be used for subcloning or exchange of immunoglobulin constant regions. Immunoglobulin expression vector allowing cloning of variable region domains and efficient purification of Fab fragments via immobilized metal affinity chromatography. Contains the immunoglobulin heavy chain region, part of the hinge region and the immunoglobulin kappa constant region. The products are secreted into the periplasm where protein folding and disulfide bond formation occurs. |
