Restriction digest of the pCAS4 gives the following sizes (kb): EcoRV--8.2,1.9.
The insert differs from that of NEB671 (ATCC 68447) by the modification the sequence encoding aa 141-143 (GATATTGAA) to GCGATCGCA. This changes the aa sequence of the protein from DIE to AIA and inactivates 3' to 5' exonuclease activity.
