Restriction digests of the clone give the following sizes (kb): BamHI--3.4; HindIII--3.0, 0.3; EcoRI--2.8, 0.5. This includes the 3' end of rat glutaminase (bp 511-1040). The coding sequence terminates 65 bp before the 3' end of the clone. In the rat, this probe detects genomic HindIII fragments of 6.0 kb, 4.4 kb, and 4.2 kb. In the human, this probe detects genomic EcoRI fragments of 7.1, 6.6, 5.5, and 4.9 kb. Only the 5.5 and 4.9 kb fragments map to chromosome 2; the others have not been localized. In the mouse, this probe detects genomic HindIII fragments of 4.8, 4.0 and 3.8 kb and genomic BamHI fragments of 22.0 and 9.4 kb. In the Chinese hamster, this probe detects genomic EcoRI fragments of 11.0, 9.2, 3.5, and 3.3 kb and genomic BamHI fragments of 25.0, 22.0, and 6.5 kb. The cRNA transcribed from pGA-1 specifically hybridizes to a 6 kb RNA present in rat kidney and brain, but not detectable in liver. This mRNA is increased 6-fold in acidotic kidneys. No RFLPs have been detected for the following strain/enzyme combinations: BALB/cHeA-STS/A (AccI, AvaI, BamHI, BglII, EcoRI, HindIII, PvuII, ScaI, TaqI, XbaI); SWR/J-C57BL/10SnJ (BamHI, EcoRI, EcoRV, HindIII, PstI, StuI, XbaI). No RFLPs have been detected for the following strain/enzyme combinations: BALB/cAnPt-DBA/2N (AccI, BamHI, BglII, EcoRI, HindIII, PstI, PvuII, TaqI, XbaI); BALB/cAnPt-C57BL/6N (AccI, BamHI, EcoRI, EcoRV, PstI, StuI, XbaI). |
