Restriction digests of the clone give the following sizes (kb): EcoRI--2.95, 1.3; BamHI--4.3; XbaI--3.7, 0.6; SacI--3.3, 0.6, 0.4. The insert contains the following restriction sites (approximate kb from the 5' end): SacI--0.4, 0.9; XhoI--.62; PvuI--.82. The insert extends from nt 163 to 1372 of the reported sequence, containing part of the coding region (nt 28-1239) and a partial 3' untranslated region. Detects mRNA of 1.7 kb in rat pineal gland. The 5' end of the insert is closer to the SK primer and the T3 promoter. Transcription from the T3 promoter gives the sense strand. |
