Restriction digests of the clone give the following sizes (kb): EcoRI--5.7; EcoRI/HindIII--3.0, 2.7; EcoRI/BamHI--5.7; BamHI--5.7; PstI--5.7; AccI--3.7, 1.45, + more; consistent with deposited DNA. Codominant segregation of the MspI polymorphism has been observed in 19 3-generation families. TaqI also reveals a polymorphism, but this has not been confirmed by pedigree studies. The probe should be pre-hybridized with excess human DNA before hybridization. Otherwise, RFLPs are observed under normal hybridization and wash stringencies.
