Restriction digests of the clone give the following sizes (kb): XbaI/XhoI--7.2, 3.2; SmaI--10.0; XbaI--10.0. A shuttle expression vector containing the DEX4 glucoamylase (GA) variant of Saccharomyces cerevisiae or S. diastaticus that encodes a sporulation-specific GA. The DEX4 signal sequence is very useful in the production of secreted fusion proteins. To use pGM30 as a secretion expression vector, remove the 2.7 kb XhoI/XbaI fragment (which contains all of DEX4 except for the upstream signal sequence). The replacement insert should have a 5' XhoI end and a 3' XbaI end for proper orientation. When transformed into yeast strain RD61-1D, pGM30 yielded approximately 70 units of glucoamylase activity. |
