Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--3.8, 2.75, 1.5; BamHI--6.6, 1.4; SalI--8.0. The insert contains the following restriction sites (approximate kb from the 5' end): BamHI--1.3, 2.6; HindIII--2.7; PvuII--0.6, 1.6; SstI--0.4, 1.7, 2.3; SalI--1.3. Two errors in the sequence of the insert have been reported. Because of a base change, the lysine coded for at nt 1351 becomes threonine and a missing cytosine at nt 1930 causes an early truncation. Digestion with EcoRI and HindIII excises the insert in two fragments. The larger fragment contains the complete coding sequence, that extends approximately from nt 60 to nt 2300. |
