Restriction digests of the clone give the following sizes (kb): SalI/BamHI--3.2, 1.1; BamHI--4.1. A NlaIII fragment extending from nt 213 to nt 1303. The NlaIII fragment was cloned into the SmaI site of pUC18. The SstI-(T4 polI)-BamHI fragment was isolated and inserted into pspT19. The plasmid can be linearized with SalI for SP6 transcription in vitro. |
