The insert contains the following unique sites in nucleotides from the 5' end: KpnI--931, XbaI--1148, HindIII--1283. Restriction digests of the clone give the following sizes (kb): BamHI--4.8; HindIII--3.6, 1.2; PstI--3.0, 1.8; EcoRI--4.8; XbaI/XhoI--3.0, 1.1, 0.7. At the 5' end, the insert includes poly(G)11 and 12 5' untranslated nucleotides. The insert contains a complete open reading frame (nucleotides 23 - 1148), and extends to the PstI site at 1848 just 5' to the poly(A). The 3' untranslated region contains 2 inverted repeat sequences and a consensus 'TATA' transcription initiation signal. The insert is oriented such that transcription from the T7 promoter gives the sense strand. |
