Restriction digests of the clone give the following sizes (kb): XhoI--10.2; EcoRI--6.3, 2.4, 1.5; BglI--5.8, 2.35, 2.05; HindIII--6.4, 3.8; BamHI--10.2. The insert contains the following restriction sites (approximate kb from the 5' end): BamHI--3.33; BglI--5.2; EcoRI--0.27, 2.68, 4.25; HindIII--1.45, 5.08; PvuI--1.35; PvuII--2.26, 5.23; SacI--2.14, 6.14, 6.15. Clone is unstable; a single colony should be picked for propagation. Constructed from pTMV204 by replacing the lambda promoter with the SP6 promoter. Linearizing with KpnI and transcription from the SP6 promoter gives the full-length, infective sense strand of the tobacco mosaic virus (TMV) strain U1. |
