The insert contains BamHI sites at positions 315 and 471 of the published sequence. Restriction digests of the clone give the following sizes (kb): EcoRI--4.2; AvaI--3.1, 1.2; BamHI--4.1, 0.2; KpnI--4.2; XbaI--4.2. The insert extends from about position 248 to the AflII site at 1294 of the published sequence. The coding sequence extends form 96 to 1295. This partial cDNA encodes part of the extracellular and all of the transmembrane and intracellular domains. The insert detects mRNA of 8.0 kb and 2.3 kb in K562, Jurkat, HL-60, and U937 cell lines. The HincII site of the vector and the AflII site of the insert were destroyed in cloning. The EcoRI site is at the 5' end. Excision with AvaI cuts at the SmaI and XhoI sites of the polylinker. |
